Cover: Schizosaccharomyces pombe and Saccharomyces cerevisiae imaged by ultrastructure expansion microscopy (U-ExM). Fourfold isotropic expansion allows the visualization of structures below the standard resolution limit of optical microscopy using conventional confocal microscopes. In S. cerevisiae, NHS ester pan-labelling (red) highlights organelles like the mitochondria and nucleus, overlaid by antibody labelling for the mitochondrial protein Cox4 (cyan). In S. pombe, the NHS ester (cyan) stains mitochondria and the nucleus more intensely, while the membrane stain Bodipy™ TR ceramide (red) provides information on vacuoles and the endoplasmic reticulum. A theory of rapid evolutionary change explaining the de novo appearance of megakaryocytes and platelets in mammals
Summary: Platelets are unique to mammals and produced from large polyploid megakaryocytes. Here, we hypothesize that platelets might have evolved because of a single event that led to the megakaryocyte polyploidization in a mammalian ancestor.

Ultrastructure expansion microscopy reveals the cellular architecture of budding and fission yeast
Summary: The resolution limit of visible light has limited ultrastructure imaging in microbes. We adapt U-ExM for budding and fission yeast, thereby improving the accessibility of super-resolution microscopy of these powerful genetic models.
